RE: [Histonet] Histosearch/Help with bone HIER

From:"Patsy Ruegg"

Yes I used 5% elmers glue coated slides for really difficult bone section
adherence but for IHC you have to be careful depending on which marker you
are using there may be non-specific binding to the glue (it is especially
the case with collagen markers as you can imagine).

You can make up a 5% solution from titebond II (glue) purchased from any
hardware store. I use it for my bone sections but I know the pathology
supervisor borrowed some of my solution for H & E's and immuno's and has had
excellent results with the tissue staying on. It is worth a try. Feel free
to contact me if you have any questions. Good luck.

I place the slides in NBF for 30 minutes prior to HIER. This seems to
help keep the tissue from falling off. Hope this helps your procedure,
to. Barb

Is anyone else having trouble searching the archives today? It keeps
freezing up on me. (It's only 51F in Chicago today - hmmmmm)

I was looking for some tips on how to keep #?+$% FFPE bone from falling

off the slide during HIER.

Tibiae are decalcifed in 5% formic acid, cut at 5 microns, dried flat
37C overnight. The marrow stays on but I'm losing the bone.

I know someone has a sure-fire answer.
