fish eggs

From:Jonathan Wilson

Hello,
A colleague of mine is going to fix some fish (trout) eggs at different
stages including hatched larvae. We tried this a while ago using Bouin's
solution and paraffin embedding using a processor but the yolk didn't
infiltrate properly in a number of eggs and yolk sac larvae. The result was
a dried up yolk ball inside the chorion or else the yolk material shattered
(very brittle). I recalled a recent tread on fixing fish embryos for
cryosectioning but was unable to find it in the archives. Could some one
with experience please offer some suggestions on fixatives and processing
schedules for paraffin.I am planning on doing some IHC on the material.
Thanks in advance.
Sincerely,
Jonathan
