Here is the method we use for Thymidylate Synthase.
Thymidylate Synthase ;
clone: TS106 (mouse IgG1-kappa)
- Mfgr/Vendor: Chemicon # MAB4130
- Control: Tonsil, known-positive colon CA, known-positive breast CA....
- Cellular localization: nuclear and cytoplasmic
- Pretreatment: 10mM Citrate solution inside a microwave pressure cooker and cooked at pressure for 15 minutes followed by a 10 minute cooldown. Wash in buffer.
- Dilution: Apply at 1:50 and incubate for 40 minutes @RT.
- Secondary: NA
- Detection: Apply EnVision+TM , DAKO Kit # K4007 (Mouse) and incubate 30 minutes @RT. Wash as above
- Chromogen: DAB
This antibody recognizes a 36 kDa protein identified as Thymidylate Synthase (TS), which is a target for the fluoropyrimidine group of anti-neoplastic drugs used to treat solid tumors. Thymidylate synthase is not restricted to a single organ system, and is present in both normal and neoplastic tissues. High levels of TS have a direct correlation with high proliferation rates.
This antibody is present in varying degrees and levels within cells that are undergoing active DNA synthesis and or repair, and is seen in both normal and neoplastic tissue. No reactivity has been observed with other cellular components.
Tumors: Expression of Thymidylate Synthase is associated with response to 5-fluorouracil (5-FU) in human breast, colorectal, gastric, head and neck carcinomas. High levels of TS in the metastases of patients with advanced disease predict lack of response to TS inhibitors such as 5-FU. High levels of TS have a direct correlation with high proliferation rates.
