Hi Ronnie,

Thanks for your input on this issue. I suspect that although I was assured
that silanised slides were used, in truth,  they were  not. I will, however,
try some of the suggestions (acetone fixation) even though this is not
advocated by all.

We have used Shadon SuperFrost Plus for all our muscle immunohistochemistry
with no problems in section retention. Yes, fat and excessive connective
tissue can be a pathological consequence of some of the muscular
dystrophies, but I haven't found this to affect section adherance. What
thickness are you cutting your sections? Usually 5-6 microns for most of our
work.
I'm assuming you are allowing your sections to completely dry before
commencing the immuno? That could leave to floating problems.

We are now using Fisher Premium Capillary Gap Plus microscope slides (100
micron gap) for all our muscle immunos, as we are now using the Microprobe
system, and getting LM results within 25 minutes, and fluorescence within 1
hour. Great results.

If I can help with anything else on the muscle, neuropath or orthopedic
front don't hesitate to ask; that's my speciality.

Although we are in the States, our lab is staffed by two Scots, one
Durbanite, and one Filipino.
