I can't speak on BioCare's antibody since we use DAKO's Melan-A (MART-1) mAb, but we get excellent immunoreactivity with regular citrate buffer retrieval in a steamer or waterbath. What tissue(s) are you staining? Not all melanomas show immunoreactivity in my experience.
