It is well known that brain tissue is the main cause of laboratory blues! No
matter which size, origin and fixative, it certainly can drive you crazy.
I have worked quite a lot with PFA-perfused rat brain, I do not know if my
method will help you, but you might give it a try.
After perfusion with saline and then 4% PFA, I fix the slice (around
5mm.)1-4 hours in the fridge (bring cold! PFA to the stables - I transport
the bucket in ice). I do not use cryoprotection, just wipe off excess liquid
after fixation, pour over some OTC (or whatever cryo mounting media you
prefer)and freeze it in n-hexane placed in a cylinder in liquid nitrogen.
