Cryoprotect!!!
20%sucrose in PBS works well in chick heart muscle. The intact heart is
not as big as what you dal with, but getting the sucrose in there really
helps. I also then infuse with a1:1mixture of the sucrose and OCT before
embedding in either OCT or the surcors/OCT mixture (see Barthel & Raymond J
Histochem Cytochem 38(9)p1383).
