Hi 'netters,

I've been trying to get calretinin (Cell Marque, polyclonal) to work on
acetone-fixed cytospins of normal mesothelial cells and known mesothelioma
cells. The antibody works beautifully on formalin-fixed, paraffin-embedded
(FFPE) mesotheliomas after HIER but I get zip on the cytology specimens. Is
this a problem with cell permeabilization? Would fixation in Zamboni's
fixative or the 3-step formalin/methanol/acetone fixation that works for ER
and PR on frozens work? Our cytospins are routinely fixed in acetone so I'm
wondering if there is some type of post-fixation step I could employ.

Any help would be greatly appreciated.

Thanks,
Linda
