mounting media bone slides

From:Linda Jenkins <jlinda@ces.clemson.edu>

Hi, Nancy,
I routinely use a drop of immersion oil or microtome oil for
temporary coverslipping of slides embedded in Technovit 7200. You need to
view, take pictures, etc. rather quickly (no longer than 1/2 day) or the
oil will start to leach some of the stain. Remove the coverslip, wipe with
a Kimwipe or equivalent tissue(do not use gauze as it has cotton fibers
that will polarize on top of your slide) and store slide in the dark or the
Technovit 7200 will continue to polymerize causing major cracks in your
section.
The oil is messy to work with but has a lovely refractive index for
close-up viewing and pictures. Whatever you do, do not coverslip with a
permanent mounting media (major cracks). Although I have comtemplated
trying to paint the surface with one of the new "hard - nail" clear
fingernail polishes. These are primarily comprised of acrylic (i.e.,
methylmethacrylate). If you have some extra slides to play with, you might
want to try this and, if you do, please let me know if it works.
	Linda
