My name is Ellen & I am a student in Zoology, The Uni. of Melbourne,Victoria Australia & am hoping someone out there can advise me with the technique for quantifying levels of secretion between glands of the uterine epithelium.
I have slides of uterine glandular epithelium (from Brushtail Possums) which can be divided into two regions, basal and superficial. The glands in these two regions secrete a particular substance (marsupial shell membrane) in different amounts, ie. the superficial glands will secrete it out of every cell, whereas the basal glands will only have 50% of cells secreting. I wish to quantify these differences so I can say that during x stage the superficial region is significantly more secretory than basal layer, but also compare the levels of secretion in each region of the epithelium between the various stages of the oestrous cycle. The amount of secretion can be seen by the redness of the glands after staining with Massons trichrome, however I would appreciate some advice as to the best method to quantify this difference between the regions and different slides.
