Oil Red O -Reply

I assume you are trying to use Oil Red O to demonstrate lipid or
lipid-containing structures in frozen sections? If so then the Oil Red O
stain can be found in most standard Hist texts.

If the Oil Red O is already in the tissue (ie perfused??) then freeze the
tissue, cut frozen sections, place on slides, airdry, rinse in water and
stain section in Haematoxylin (if desired), coverslip in an aqueous
mountant. You could fix the sections in formalin for 10 min or so prior to
coverslipping. Any structures containing the Oil Red O will appear red. I
can't imagine why this type of technique would be done and whether
perusion with Oil Red O would actually work. Pleas supply more info.

Can anyone provide me with a protocol for
demonstrating Oil red O on a fresh, unfixed tissue
section? Is this technique possible?
