Dear all: The best way to croprote ct brain tissue of any organism is to
start at 10%-on a stir plate in a cold room overnight---or until it
sinks----then go into 20% sucrose until that sinks----then into 30%
sucrose. You must use gradients of sucrose for brain. Best to have on a
stir plate with stir bar in the COLD. I have done that for years.
Just pull out of 30% and place into 10%---if it sinks place in the 20%
for at least two days--probably. Then go into fresh 30% sucrose.
