Re: [Histonet] Hematoxylin-eosin-saffron staining

From:Rene J Buesa

Hi Luminita:
  There is a procedure by Langeron (1942) for saffron and eosin B:
  Sol.A = 1% eosin; Sol.B = dist.water¡ú100mL + saffron¡ú2 g + + 5% aq. tanin¡ú1 mL + 40% formaldehyde¡ú 1 mL
  Procedure:
  hydrated sections stained with hematoxylin ¡údifferentiated ¡ú nuclei blue ¡ú sol A for 5 min¡ú quick rinse in water ¡ú 70% ethanol for a few seconds ¡ú wash well in water ¡ú sol B for 5 min¡ú BLOT sections ¡ú flood the sections with absolute ethanol until dehydrated ¡ú xylene ¡ú cover slip.
  For the macrophages I would stain toluidine blue.
  Hope this will help.

  Hi,

I am looking to find the protocol for the hematoxylin-eosin-saffron
method and I have difficulties. I would like to ask if anybody can share
with me this protocol?. I appreciated so much. Also, does somebody knows
a staining for macrophages from the lung or synovial tissue , using only
histochemistry staining without immunostaining?. Thank you again.

Sincerely,
Luminita
