Hi all and happy new year!

I have muscle tissue sections stained for gelatinase enzyme activity using a
fluorescent marker. I have images from 27 samples and a negative control
image (no substrate) for each. I want to quantitatively analyse the images
for both intensity and area of signal. Has anyone done this sort of image
analysis before or know of a gold standard reference for it?
I'm not sure whether to use a thresholding selection approach or look at the
raw data from an intensity histogram for both negative control and sample
and only count pixels which do not overlap in intensity values......
