For urgent frozen sections we used to sit the tissue in tissuetek on the cryostat chuck and just dunk it in liquid nitrogen. The sections on the slides were then post fixed briefly in acetone and stained with a rapid H&E. This was good enough for a quick diagnosis, resection lines etc. BUT the blocks were always then processed in the usual manner afterwards and re-examined by the pathologist.


I can send you methods for fixation/cryoprotection for rat tissue if you would like. What are you going to do with the tissue for your experiment?

     Does anyone have a protocol on freezing tissue in liquid nitrogen?  I know
its usually best to fix the tissue, then cryoprotect it.  I have a professor
that just want to freeze the tissue without a fixative and cryoprotection?
       
     Also, I need a protocol on fixation/cryoprotection for another
experiment.  Any help would be appreciated..
