Happy Chinese New Year to all in Histoland.
I am doing fresh frozen sections of muscle and am getting freezing artifact.
My protocol is to remove the muscle and embed in OCT in a CO2/ methylbutane
slurry. Then I section at -18, fix the sections, and b-Gal stain. My
question is this: can I cryo-protect with sucrose prior to
embedding/freezing without fixing the muscle first? Would the sucrose
interfere with staining?
