Vacuoles Artifacts

Help please Histonetters,
Occasionally experiencing small vacuoles in the epidermis of skin processed
for Mohs. Today since the Dr. was processing a small melanoma case it was
esp. aggravating. Any ideas on what causes this and how to rid the finished
product of this confusing artifact?? I do frozen sections in -22 C to -25 C
cryostat chamber-have never used liquid Nitrogen and probably won't start
now. Do have access to a -45 C cold bar in a second cryostat-doesn't appear
to be a staining phenomena but more a processing fluke. Any ideas will be
greatly appreciated.
