Denise,

Ideally Cut frozens, fix in absolute alcohol and then use your frozen
section staining set-up to stain.

Or you can use alcohol-fixed, paraffin-embedded tissue. once it's fixed in
alcohol the urate crystals will not leach out.

See:
Histotechnology, a self-instructional text, Carson, 2nd Ed p. 208, 219.
or
Theory and Practice of Hitological Techniques, Bancroft & Stevens, 4th ed.,
p.260

Subject: two questions

Hi gang! Two questions for today:
1. does anyone have a recipe/procedure for a totally alcoholic
hematoxylin? Some colleagues have a gout specimen and don't want the
crystals to dissolve out, but need an H&E.
2. has anyone tried the modified Crossman's trichrome procedure
recently published in the Journal of Histotechnology?
If so, how did it work?
Many, many thanks in advance for all your input. (This histonet is just
SLICK!)
