I've used the same protocol (R.P. Bird) quite often and with good
results. In the beginning the staining was too strong so I reduced the
concentration to 0.1% dissolved in PBS or even methanol. And the maximum
time of staining was at best about 7 minutes. After dissolving the
methylene blue I filtered this solution before I used it.

Does anyone have experience staining rat intestine with methylene blue
to look for aberrent dysplastic crypts. The mucosal side is examined
directly under a dissecting microscope, the method is that of RP Bird
from Cancer Letters 37 (1987) 147-151
I am having difficulty differentiating the crypts as everything stains
intensely
