Zamboni-fix
From: | Kenneth Wester <kenneth.wester@genpat.uu.se> |
Hi All.
Is there anyone who can give me informatiom on how to properly use the
Zamboni-fixative on cryo-sections? In my hands, the immunoreactivity is
preserved excellently, e.g., for EGF-receptor staining, after Zamboni
fiaxation. However, the overall morphology is poor, even when compared to
fixation in aceton and/or methanol.
I have tested air-drying the sections for between 30 seconds up to 1 hour,
without any major improvement. Usually I fix for 2-3 minutes followed by
rinsing in distilled water.
Greatful for any tips regarding the morphology problem,
Kenneth Wester
