Dear Ty Lee,
The antibody I used, a rabbit polyclonal cat no. A3537, reacts with the 32kD
proenzyme with minor bands demonstrated by immunoblot of tissue extract. The
minor bands are thought to result from proteolytic degradation rather than
activation. The 38kD band found on Western blots of liver and kidney may be
the result of alternative splicing, or post-translational modification in
these tissues. No reactivity with the two subunits was found. This
information is taken from Dako's data sheet.
Apparently Promega has an antibody specific for the activated enzyme. See
Brett Connolly's post on Histonet today.
