Hi Vicki,
I didn't experience this with the NexES, but I did
have some of the same problems with the ES.
Amplification isn't necessary with NSE or Melanosome
(I've had experience with both Dako & Ventana
clones/antibodies). I did use a 3% H2O2 in METOH
block for 10 minutes prior to starting the program
though. The NSE is especially prone to non-specific
staining in the control because pancreas autolyzes
(did I spell that right? It's after 10pm and my brain
is ready for bed) and that picks up like iron filing
to a magnet.
