Hi Taffy,
I found the article "Detection of Apoptosis by a Modified Trichrome
Technique," in the Dec. 1999, Journal of Histotechnology, vol. 22, No.4.
It over stained my bone sections in parapffin badly so I did
further modification and ended up with the staining procedure below:
3 seconds in Acid fuchsin 0.50 gm
Orange G 0.025 gm
Glacial acetic 1 ml
acid
Distilled H20 up to 100 ml
Rinse in DH20
1 minute 5% phosphotungstic acid in DH20
Rinse in DH20
3 minutes in Light Green .50 gm
Glacial acetic 1 ml
acid
Distilled H20 up to 100 ml
Rinse in DH20
Brief rinse in 100% ETOH
3 changes Clearing Agent
I have not done any specimens in plastic, but this works well for the
paraffin sections. Hope it works for you.
