I cut a lot a mouse tissue and some of it is pretty old and dried out. I
soak blocks minimally in ice water nothing added, float for as long as
possible, use charged slides and dry overnight at room temperature. I have
steamed, autoclaved and microwaved sections will little or no loss. Are the
sections comming completely off the slides? In my hands greater than 0.3%
H202 can cause tissue destruction and the use of coverslips during staining
can also be a problem. Have also found that boiling during HIER is
detrimental. Hope this helps.
