does anyone know what may be causing my frozen mouse brain sections to
become detached from the gum tragcanth cork mounting upon placing them in
an ultralow freezer for holding/storage ? We freeze our tissue samples
mounted on the cork with the gum tragcanth and immerse in isopentane
chilled by liquid nitrogen for 12 sec. and then we store them at -80C until
sectioning. Up until this point I haven't had any problems. The majority
of our tissuses have been muscle ( cat/dog) and for some reason these
mouse tissues are driving me crazy especially the brain. Any and all help
will be much appreciated. Thanks, Atoska
