--We use cold acetone to fix our murine hearts for immunos. 5 minutes in the
cryostat. Silane treated slides. (green slide holders with LIDS, acetone is
very volatile, moderately toxic if inhaled and even narcotic) the samples
are never really air dried before fixation, just what they get from
"melting" to the slide. They immediately go into acetone. When I take the
slides out of acetone the "fixed" OCT surrounding the tissue peels off
leaving the tissue. Then the slide is air dried a couple of seconds and
filed for later use. Beautiful cryosections and immunos with Has2
antibodies. anita
