I have a question about the DAKO Catalyzed Signal Amplification System,
Peroxidase. I 've tried to use it on frozen sections, but whatever I do
there is a tremendous background staining. Even in the control sections
(=negative control reagent instead of primary antibody) everything is
stained. The sections are fixed in formalin and I 've done biotin and
peroxidase blocking. For wash buffer I 've used PBS + 0.1% Tween 20. I want
to use the kit for antibodies that work well on frozen sections (most of
the time rat liver sections), but are a little bit faint. Is it possible to
use it on frozen sections or is the kit only for paraffin sections? Thanks!
