Richard,
We use Direct Red 80 from Sigma Chem. Co. (Cat# D 030) in our Picrosirius
Red staining method for collagen. You view your PS Red stained slides under
cross-polarized light to view the birefringent collagen. This can be
quantitated using image analysis. The PS Red method stains mostly collagen
types I, II, and III.
PS Red Staining solution:
0.25 g Direct Red 80 (Sirius Red F3B)
in
250 ml Saturated Picric Acid (Sigma Chem. Co)
mix and filter
We follow the staining method by Junquera et al.
