We tried NF-kB staining in mice frozen heart tissue today. We did not
get staining inside myocyte at all. (none in nuclei, cytosol). We used
santa cruz 109 and 372 antibody against p65. We only get fluorescense
in intercellular space and membrane.

Are there anybody who tried theses antibody?
We just blocked with normal goat serum for 1 hour before the primary
antibody.
Do you think we should make more hole in cell membrane, or should we
employ antigen retrieval method?

please give me some advice.
