Peter,

You may find that the staining corresponds to the nucleolus. I have found
that with many mRNA-ISH reactions, if there is cytoplasmic staining then
there is often nucleoloar staining as well. Not surprising since mRNA is
initially produced in the nucleus. I will upload to Histonet some pics on
this type of staining shortly.

Hello Histonetters,

I am currently doing in-situ hybridization on paraffin embedded tissue array
sections.  Very often I get a signal in the nucleus, which I think is a
little odd as I would expect to see it in the cytoplasm.  
Has anybody else encounted this?
If so, is it real?
If it isn't real, what can be done about it?
