In the veterinary field I routinely have to do this when working on certain
lesions/infections (especially those that have a lot of free Igs in the
tissues).  I just add 2% normal serum from the case species to the secondary
antibody per volume.  It helps to block 'nonspecific' binding of the
secondary to Igs in the tissues.  It does not appear to affect the specific
binding of the secondary to the primary rb/ms Ab.  I would assume it would
work with human serum as a blocker as well.
