Re: Bodians stain

From:Nancy E Weber

Hi Ray,
Here's a Churukian Schenk that I used with bodian's.

Fixative: 10% neutral buffered formalin
Technique: Cut paraffin sections at 3 to 5 microns
Quality Control: An argyrophil-positive carcinoid tumor is preferred, but a
section of small intestive can be used.

Staining Procedure:

1. Deparaffinize and hydrate sections to acidified Water, ph 4.2.
2. Place slides in 0.5% silver nitrate and heat in microwave on "warm" for
2.5 min.
3. Rinse in 3 changes of warm distilled water.
4. Heat Bodian's developer in microwave on "warm" for 2.5 min. Place slides
in developer for 3 mins.
5. Rinse in 3 changes of warmed distilled water.
6. wash gently in running tap water.
7. Rinse in 3 changes of warmed distilled water.
8. Heat same silver nitrate solution as in step 2, on "warm" for approx. 2
mins. Place slides in warmed silver solution for 5 min.
9. Rinse in 3 changes of warmed distilled water.
10. Heat same Bodian's developer as in step 4 on "warm" for approx. 2 mins.
Place slikdes in warmed developer for 3 min.
11 Rinse in 3 changes of warmed distilled water.
12. Counterstain with 0.1% nuclear fast red for 3 min.
13. Rinse in distilled water.
14. Dehydrate, clear, and mount.

Results:
     Argyrophil cells.....................................black
     Argentaffin substances..........................black
     Nuclei.....................................................red
     Backround...................................yellow-brown

Solutions:

0.3% Citric Acid
    Citric acid.............................................0.3 gm
    distilled water.....................................100.0 ml

Acidified water
     Distilled water.....................................500.0ml
Ph to 4.2 with 0.3% citric acid sol'n.

0.5% Silver Nitrate
    Silver Nitrate........................................0.5 gm
    Acidified water.....................................100.0 ml

Bodian's Developer
     Sodium Sulfite (anhydrous).....................5.0 gm
     Hydroquinone........................................1.0 gm
     Distilled water....................................100.0 ml

Nuclear Fast Red (Kernechtrot)
     Nuclear Fast Red..................................0.1 gm
     Aluminum Sulfate..................................5.0 gm
     Distilled water.....................................100.0 ml
Dissolve aluminum sulfate in distilled water, then add nuclear fast red and
dissolve with aid of gentle heat. Cool, filter and add a grain of thymol
 as a preservative.
