RE: RISH In Situ counter stain

From:Cynthia Favara

Mary,

I have no experience with your technique. I think you are doing some sort of
InSitu?
I have had signals masked in IHC when counterstaining with Harris and
sometimes will even leave off the counterstain depending on what we are
looking for. I have had success with decolorizing hematoxylin with a dilute
<1%HCL in 70%ETOH provided the markers will with stand that.

Good luck and enjoy your Friday!

Subject: RISH In Situ counterstain

Good Morning,
Has anyone had experience with counterstaining RISH slides with Harris
hematoxylin rather than Mayers? A long sad story but now I find that I have
masked signal and am wondering if it could be the Harris. Any wonderful
suggestions? I am wondering if I can take the coverslips off without
compromising the emulsion and destain in dilute HCl.
Thank you in advance for any pearls of wisdom!
