processing plant seeds
From: | Andrea Grantham (by way of Histonet) |
I posted this yesterday AM and since then have had no histonet messages.
Forgive me if this is a repeat - I'm not sure that the absence of histonet
messages has something to do with histonet or the system here (which has
been flaky in the last week). So I try again...I have some of the seeds now
and need to get them processed.

Yesterday's post:
This week I will start receiving Guayule seeds for processing and
sectioning. Having never done seeds before I'm hoping that there may be
some plant histologists out there who can impart some advice. I do have a
paper with a protocol for processing using FAA to fix the seeds and ETOH
and Xylene in the processing - the whole procedure takes about 5-6 days to
complete.
There was a mention in the paper of using butanol in the processing - is
this a better choice for dehydrating and clearing?
The investigator also references a book, Plant Cell Biology, 2nd edition by
Jackie Spence and I am borrowing a copy which should arrive this afternoon.
Is there another source of information on processing seeds?
Thanks in advance!!

One other question - would it be OK to replace xylene with Clear-Rite 3?
