hier on bone sections

a couple of years ago we stopped doing any hier on our bone sections because
of losing the sections, we seemed to mostly lose cartilage selectively and
that was just what we were interested in.  first, we switched to zinc
formalin fixation which illiminated the need for AR in most cases and when
we do need AR we use enzyme digestion which we feel we can control better
than hier.  we use proteinase k in most cases but sometimes have to use
chondroitinase-hyaluronidase-pepsin-keretinase combinations not so much to
access crosslinked proteins from fixation but to digest thru the molecule
segments to get at the antigen of interest.
