One thing to add to the GFP discussion - you CAN do multiple GFP
expressions, for example, with YFP (yellow) and CFP (cyan), but you have
to be certain that your filters are tight enough. I don't have any
experience with paraffin processing of GFP, but I am finding that
immunodetection at the TEM level seems to be construct-dependent. Which
may just be me :), but it is VERY frustrating! Anyway, that may have some
bearing on the paraffin-processing variability; some constructs or mutants
of wild-type GFP may withstand fixation/dehydration better than others.
