Hello All,
My Factor VIII protocol is now working on control tissues thanks to 
your help in suggesting the enzyme digestion step. However, the 
tissues from the study are still not staining. They are sections of 
mouse skin (FFPE-4 um). I suspect they were fixed for up to two weeks 
before processing. 
Questions:   a) Am correct to think fixation is a likely cause of 
                           the lack of positive reaction?

                        b) Is there something I can try to overcome 
                            this problem? 
 I am using trypsin for 10  mins. at 37 C. 
                         c) Should I try 20 mins? 
                         d) Would a different enzyme make a 
                              difference?
