I use 10% serum of the species from which the 2nd is made, 5% RIA grade
BSA in buffer (TRIS or PBS). Depending on the specific antibody, I may
include 0.1% Triton X-100 or Tween 20. The 10% normal serum is probably
overkill since when I double label in a cocktail the sera are at 5%.
I think this is the heart of the basic recipe. Variants derive from what
lab you grew up in and what specifics you're dealing with. Or so I think.
