We are doing tunnel. We have done caspase 3 and it does not correlate with
tunnel.

You could look at performing tunel in which you would need to purchase a kit
or you could stain immunohistochemically for cleaved caspase 3, which is a
marker of apoptosis. The heat should not affect the detection of apoptosis
in fixed tissue. To induce apoptosis in cell culture you can place the
cells at 36C for 30 minutes, but you need to control the temp.

I need a protocol for apoptosis for paraffin embedded tissue. Has incubating

paraffin sections at 50 degrees for a certain length of time any effect on

the out come of the apoptosis staining? Thanks.
