in situ zymography

From:NIDAL E MUVARAK

Hi everyone. I'm want to perform an in situ zymography assay on mouse lung tissue. This will be the first time I'm doing this kind of assay. I have a protocol my PI used at her previous institution, however , she mentioned that it didn't work for her. So I was wondering if anyone out there has a protocol for in situ zymography, or has any suggestions on some gelatinase activity kits. Briefly, I harvest the left lung from the mouse and perfuse it through the pulmonary artery with OCT/PBS (50:50). Then freeze it in OCT for sectioning. After that, for whatever assay I'm doing, I fix in cold aceton. Thank you for your time.
