ISH probe for whole mount vs slide staining
We have some ISH riboprobe that has been used successfully on stage 11 chick embryos for whole mount techniques, and want to use it on cryosections. What, if any, changes to the probe concentration or hybridization time/temperature, etc. do we need to make? Or should it work just as well at the same concentration and hybridization time and temperature? We have tried this twice, once with one probe and got too much staining, and one with a different probe with no staining. Before we try this again, I would like to borrow from your collective wisdom...
