Hi all,

I am preparing mouse embryo in gelatin for vibratome and cryostat sectioning. Are there any differences in preparing the two gelatin molds for vibratome and for cryostat sectioning? I prepared the embryo in gelatin for cryo sections the same way I prepared the other embryo for vibratome sectioning and I had many holes in the embryo sections as the same type of holes in the outer part of the gelatin section. All protocols would be appreciated as I am working with ISH stained and non ISH stained mouse embryo, if this makes a difference.

Thanks!
