Re: Loyez procedure
Nobody else has posted an answer to this one, so
here you are. The method is for paraffin sections
of formaldehyde-fixed CNS.
1. Treat hydrated sections with 4% iron alum, overnight, room
temp.
2. Wash in water for 5 min.
3. Stain overnight, room temp, in a freshly prepared mixture of:
10% alcoholic haematoxylin (well ripened) 10 ml
Water 90 ml
Sat. aq. lithium carbonate 4 ml
(Alternatively 3 hours at 56C)
4. Wash in water.
5. Immerse in 4% iron alum to remove much of the background
colour.
6. Wash in water.
7. Immerse in borax-ferricyanide (1% borax, 1.25% K3Fe(CN)6 in
water) until only the myelin sheaths remain stained
(blue-black).
8. Wash in water; Optionally counterstain with eosin;
dehydrate, clear and cover.
