I always use the h2o2 block after the primary antibody.

To the Histonetter who was wondering wether to use the hydrogen 
peroxide before or after the antibody incubation , I use it before the 
incubation but have read that it can mask or destroy some antigen 
sites. So if you're having problems , try doing it directly after the 
incubation.
