Theory and Practice of Histotechnology, Sheehan and Hrapchak pp 94-95,
Second Edition has discussion on EDTA decalcified bone/paraffin sections/AP
staining. Although an older book, the discussion on enzyme staining of
decalcified bone for alk phos AND acid phos is excellent. This is probably
discussed in other texts/literature as well. What decalcifier used is
critical for both of these enzymes.
Did you restore alk phos enzyme activity with magnesium chloride? EDTA
decalcified bone must be treated with 1% magnesium chloride for 2 - 6 hours
or longer to restore enzyme activity.
