I'm going to be running an immuno stain for COX-2 on rat and mouse bone
fractures. I know there are a lot of you out there who run this stain, and
was looking for any suggestions that you may have. We're using a polyclonal
rabbit primary antibody to murine COX-2 (Cayman Chemical Company), a
secondary anti-rabbit conjugated to HRP, and DAB as our chromagen. I was
planning on using our normal blocking procedure: 3% hydrogen peroxide for
endogenous peroxidase, and 3% powdered milk in 1% BSA to block non-specific
sites. Is antigen retrieval recommended for COX-2? If so, what type?
Any help is greatly appreciated.
