I have done some IF work, yes leaving the antibody on too long will yield 
high background and pick up dust particles , etc. Also some flourescent 
antibodies do not need secondary or pre-treatments,third you should use a 
water soluble chromagen for one antibody, then the flourescent antibody, most 
flourescent antibodies are coverslipped with a water based mountant, not 
great for permanent
